Ayat 1 sampai 5 dari surah Al-Alaq merupakan ayat-ayat Al-Quran yang paling pertama diturunkan saat Nabi Muhammad Saw. sedang melakukan tafakur perenungan di gua Hira. Surah ini pun punya nama lain, yakni Iqra’ atau Al-Qalam. Dari benda hina tersebut, manusia terbentuk dan Allah Swt. memuliakan mereka dengan diajari cara membaca, menulis, serta diberikan ilmu pengetahuan. Namun sayangnya, manusia yang melupakan asal-usulnya malah tidak mensyukuri nikmat yang Allah berikan sampai bertindak di luar batas; Dijelaskan pula tentang ancaman dari Allah Swt. terhadap kaum kafir yang berusaha menghalangi kaum muslimin saat melaksanakan perintah dari-Nya; Surah Al-Alaq mempunyai hubungan dengan surah Al-Qadr. Jika dalam Al-Alaq dijelaskan tentang perintah membaca Al-Quran dari Allah Swt. bagi Rasulullah Saw., maka Al-Qadr memaparkan tentang proses turunnya Al-Quran.
